IMP761 is a first-in-class, humanized IgG4 monoclonal antibody that acts as an agonist of lymphocyte-activation gene-3 (LAG-3). It is designed as an immunosuppressive therapy to address the root cause of autoimmune diseases by specifically silencing self-antigen-specific memory T cells that accumulate at disease sites, thereby restoring immune balance. By enhancing the inhibitory "brake" function of LAG-3 on activated T cells, IMP761 suppresses dysregulated immune responses without broadly suppressing the entire immune system. Preclinical studies have shown that IMP761 inhibits peptide-induced T cell proliferation and activation, reduces effector cytokine production, and decreases antigen-specific delayed-type hypersensitivity reactions. Early clinical data demonstrate significant suppression of T cell activity (up to 80% inhibition at 0.9 mg/kg) with a favorable safety profile in healthy volunteers. The drug is being developed for multiple autoimmune diseases including multiple sclerosis, rheumatoid arthritis, type 1 diabetes, and oligoarticular juvenile idiopathic arthritis[1][2][6][8].
