IMP9064 is an orally bioavailable small molecule inhibitor of the DNA damage response (DDR) protein kinase ataxia telangiectasia and Rad3 related (ATR) kinase. It is designed to block ATR, a key regulator of cell cycle checkpoints and DNA repair in response to replication stress or DNA damage, thereby promoting cancer cell death through checkpoint inhibition and antineoplastic activity. Discovered and developed by IMPACT Therapeutics, IMP9064 has demonstrated high potency and selectivity for ATR over other kinases in preclinical studies, with notable activity in ATM-deficient cell lines. The drug is being evaluated as monotherapy and in combination with the PARP inhibitor senaparib for patients with advanced solid tumors[1][3][4][5].
