IMPT-601 is a bispecific chimeric antigen receptor (CAR) T-cell therapy developed by ImmPACT Bio for the treatment of gastric cancer and other solid tumors expressing Claudin 18.2 and secreting transforming growth factor beta (TGF-β). The therapy employs an "OR-gate" logic design to target both Claudin 18.2 and TGF-β, aiming to overcome the immunosuppressive tumor microenvironment by enhancing CAR T-cell infiltration, reducing regulatory T-cell activity, overcoming T-cell exhaustion, and converting the typically suppressive signal of TGF-β into a pro-inflammatory activation signal for CAR-T cells. Preclinical studies have demonstrated robust anti-tumor activity with persistent cytotoxicity against tumor cells expressing both targets, reduced exhaustion markers in CAR-T cells, decreased regulatory T cell differentiation, and improved lymphocyte infiltration into tumors compared to single-input CLDN18.2-targeting CAR-T therapies[1][3][6].
