Overview

IMT-009 is a first-in-class, humanized, Fc-attenuated IgG1 monoclonal antibody designed to inhibit CD161 (KLRB1), a C-type lectin receptor expressed on natural killer (NK) cells and memory T cells. By blocking the interaction between CD161 and its ligand CLEC2D (LLT1), IMT-009 aims to overcome immune suppression in the tumor microenvironment, enhancing NK cell degranulation and T cell-mediated killing of tumor cells. Developed by Immunitas Therapeutics using a single-cell genomics platform, the drug is being investigated for the treatment of advanced solid tumors (including microsatellite stable colorectal cancer, NSCLC, and HNSCC) and hematological malignancies (lymphomas). It is being studied both as a monotherapy and in combination with other agents like fruquintinib.
