IMT-380 is a **fully human, Fc-active anti-CD161 monoclonal antibody** designed to selectively deplete pathogenic CD161+ immune cell populations, thereby reducing tissue inflammation and restoring immune homeostasis in autoimmune settings. CD161-expressing T cell subsets have been implicated in autoimmune diseases due to their production of inflammatory cytokines (including IL-17A, IFN-γ, IL-22, TNF-α, and GM-CSF) and their presence in inflamed tissues. IMT-380 works by targeting CD161, which remains a stable marker on these pathogenic T cells, making it a precise therapeutic target. The antibody is designed with Fc-active properties to enable antibody-dependent cellular cytotoxicity (ADCC) and other effector functions for cell depletion. **Developed by Immunitas Therapeutics**, IMT-380 represents a novel targeted approach for treating autoimmune diseases as an alternative to broad immunosuppression. Preclinical data demonstrated that IMT-380 effectively depletes CD161+ T cells in vitro and in vivo, significantly reduces inflammatory cytokine production, and improves disease severity in animal models including a non-human primate (NHP) psoriasis model where it reduced psoriasis area and severity index (PASI) scores. The antibody showed high specificity and potency, with CD161+ cells being highly enriched in inflamed intestinal tissue from Crohn's disease patients.
