Overview

IMT1 is a first-in-class, highly specific and noncompetitive inhibitor of human mitochondrial RNA polymerase (POLRMT). It induces conformational changes in POLRMT, inhibiting mitochondrial transcription and consequently disrupting mitochondrial function. IMT1 exhibits potent anti-cancer effects in vitro and in vivo, notably against osteosarcoma and endometrial carcinoma cells, where it impairs cancer cell proliferation, induces apoptosis, and disrupts mitochondrial membrane potential and ATP production. IMT1 suppresses the Akt-mTOR signaling pathway in cancer cells and demonstrates selectivity for cancer cells over normal cells. It is administered orally and represents a new strategy for targeting mitochondrial transcription in cancer therapy[1][2][3][4][5][6][7][10].
