The combination of **imvotamab** and **loncastuximab tesirine** is an experimental antibody-based therapy for B-cell lymphomas, leveraging two distinct and potentially synergistic mechanisms of action. - **Imvotamab** is a bispecific (CD20 x CD3) IgM monoclonal antibody that targets CD20 on B cells and CD3 on T cells, facilitating T-cell mediated killing of CD20-expressing tumor cells with reduced risk of cytokine release syndrome and neurotoxicity compared to IgG-based T-cell engagers. - **Loncastuximab tesirine** is an FDA-approved antibody-drug conjugate (ADC) consisting of a humanized anti-CD19 antibody conjugated via a protease-cleavable dipeptide linker to a pyrrolobenzodiazepine (PBD) dimer toxin (SG3199), enabling targeted cytotoxicity against CD19-positive B cells through DNA crosslinking and induction of apoptosis. This combination is in phase 1b/2 trials for relapsed/refractory non-Hodgkin lymphomas, including diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma, based on rationale that simultaneous targeting of CD19 and CD20 may overcome resistance and improve clinical outcomes in B-cell malignancies[1][2][4].
