IN-N01 is a humanized IgG4 monoclonal antibody targeting the amino terminus of amyloid beta (Aβ), developed by Intellect Neurosciences in collaboration with MRC Technology as a next‑generation immunotherapy for Alzheimer’s disease and other amyloid-related disorders. It is derived from the mouse monoclonal antibody 82E1 and engineered to bind monomeric, oligomeric, and plaque forms of Aβ without binding amyloid precursor protein (APP), with the IgG4 isotype selected to minimize Fc-mediated inflammatory responses in the brain. Preclinical rationale is to promote clearance of toxic Aβ species while reducing neuroinflammation and edema risks associated with IgG1 anti-Aβ antibodies, positioning IN‑N01 for potential disease-modifying treatment in early or presymptomatic Alzheimer’s disease and for exploration in indications such as traumatic brain injury–associated amyloidosis and retinal amyloid deposition.
