This experimental in vivo CAR-T therapy, developed by The 923rd Hospital of Joint Logistics Support Force of People's Liberation Army, utilizes lipid nanoparticles (LNPs) to deliver mRNA encoding a CD20-specific chimeric antigen receptor (CAR) directly into the patient's body. Unlike traditional ex vivo CAR-T cell therapies, which require complex and time-consuming laboratory modification of a patient's T cells, this approach aims to reprogram T cells in situ. Once the mRNA is delivered and translated, the patient's own T cells transiently express the CAR, allowing them to identify and eliminate CD20-positive malignant B cells. This technology is being explored as a potentially more accessible, cost-effective, and rapid treatment for various B-cell hematological malignancies, including B-cell lymphoma and lymphocytic leukemia.
