This is a four-drug **experimental combination regimen** including IN10018 (a selective oral focal adhesion kinase [FAK] inhibitor), KN046 (a bispecific immune checkpoint inhibitor targeting PD-L1 and CTLA-4), albumin-bound paclitaxel (a nanoparticle, protein-bound formulation of the antimicrotubule/taxane paclitaxel, known commercially as Abraxane), and gemcitabine (a nucleoside analog antimetabolite chemotherapeutic). - **IN10018** blocks FAK, a kinase involved in signaling pathways that regulate tumor cell survival, invasion, and the tumor microenvironment[3][5][7]. - **KN046** binds to immune checkpoint proteins PD-L1 and CTLA-4 to enhance anti-tumor immune activity (dual checkpoint blockade)[3][7]. - **Albumin-bound paclitaxel** (Abraxane) disrupts microtubule dynamics, arresting cells in mitosis and triggering apoptosis, with improved tumor delivery via albumin-mediated transcytosis and SPARC binding[2][4][6][8]. - **Gemcitabine** is a deoxycytidine analog that is incorporated into DNA during replication, leading to inhibition of DNA synthesis and apoptosis. This regimen is being investigated in solid tumors, recurrent or metastatic diseases, especially where combination immunotherapy and cytotoxic chemotherapy may have synergistic effects[7][9].
