Overview

Inactivated Akkermansia muciniphila (specifically strain Akk11 in the context of Shanghai East Hospital) is a microbial therapy consisting of pasteurized or heat-killed bacteria. Unlike live biotherapeutics, the inactivated form of this gut commensal has been shown to retain or even enhance its metabolic benefits, particularly in improving intestinal barrier function and insulin sensitivity. The primary mechanism involves the interaction of the outer membrane protein Amuc_1100 with Toll-like receptor 2 (TLR2) on the intestinal epithelium. This interaction triggers signaling pathways that strengthen tight junctions and reduce metabolic endotoxemia. It is currently being investigated in Phase 4 clinical trials for the treatment of polycystic ovary syndrome (PCOS) and obesity, often as an adjunct to metformin therapy.
