Inactivated non-typeable Haemophilus influenzae (NTHi) is an oral bacterial vaccine candidate, specifically developed as **HI-164OV**, designed to prevent acute exacerbations in patients with chronic obstructive pulmonary disease (COPD) and chronic bronchitis. The vaccine consists of killed whole-cell NTHi bacteria. Its mechanism of action is based on the concept of the common mucosal immune system; oral administration stimulates gut-associated lymphoid tissue (GALT), which primes NTHi-specific T-lymphocytes. These primed cells then migrate to the bronchial mucosa, where they enhance local immune defenses and reduce bacterial colonization and mucosal inflammation. Clinical trials have indicated that this immunotherapy can reduce the frequency and severity of exacerbations and the need for antibiotic treatment in susceptible populations, such as smokers with chronic airway disease.
