Inactivated rotavirus vaccine (IRV) is a non-replicating, parenterally administered vaccine developed to prevent rotavirus gastroenteritis. Unlike currently licensed oral live-attenuated vaccines, IRVs are produced by chemically or physically inactivating whole rotavirus particles using agents such as formaldehyde, beta-propiolactone, binary ethyleneimine, heat treatment, or ultraviolet irradiation. These methods aim to preserve the antigenic structure of the virus while eliminating infectivity. Preclinical studies have shown that IRVs can induce strong serum antibody responses and mucosal immunity when administered intramuscularly or via microneedle patches. The primary indication for this vaccine is prevention of severe diarrhea caused by rotavirus infection in infants and young children[4][5]. The development of IRV seeks to address safety concerns associated with live oral vaccines and improve efficacy in populations where current vaccines underperform.
