Overview

Inactivated trivalent influenza vaccine (TIV) is a seasonal biological preparation designed to provide immunity against three distinct strains of the influenza virus: typically two influenza A subtypes (H1N1 and H3N2) and one influenza B lineage (either Victoria or Yamagata). The vaccine is produced by growing the viruses in embryonated chicken eggs or cell culture, followed by inactivation (killing) using chemical agents such as formaldehyde. The primary mechanism of action involves the induction of neutralizing antibodies against the viral hemagglutinin (HA) protein, which prevents the virus from attaching to and entering host respiratory epithelial cells. TIVs have been the standard of care for seasonal influenza prevention for decades, although they are increasingly being replaced by quadrivalent formulations (QIV) that include an additional B-lineage strain. In clinical research, TIV is frequently used to evaluate vaccine immunogenicity and safety in specific populations, such as patients receiving immune checkpoint inhibitors or those with autoimmune conditions.
