V212 is an investigational, heat-treated (inactivated) varicella-zoster virus (VZV) vaccine developed for the prevention of herpes zoster (shingles), particularly in immunocompromised individuals. The mechanism of action involves stimulating the immune system to generate specific anti-VZV antibodies and cell-mediated immunity against the varicella-zoster virus. Developed by Merck, V212 was designed as a non-live alternative to existing shingles vaccines for patients at higher risk due to immunosuppression. Clinical trials demonstrated that V212 reduced the incidence of confirmed herpes zoster cases by approximately 64% and also decreased complications such as post-herpetic neuralgia and hospitalizations related to herpes zoster. Despite promising results in phase 3 trials, development for this indication has been discontinued[1][2][3][4].
