Overview

Inarigivir soproxil is a first-in-class oral small molecule antiviral drug developed as a prodrug of SB 9000. It acts as an agonist of innate immunity by selectively activating the retinoic acid-inducible gene I (RIG-I, also known as DDX58) and nucleotide-binding oligomerization domain-containing protein 2 (NOD2) pathways within infected cells. This activation induces the interferon signaling cascade, enhancing antiviral defense mechanisms and inhibiting viral replication by interfering with hepatitis B virus (HBV) polymerase and pregenomic RNA interaction. Inarigivir was primarily investigated for chronic hepatitis B infection but has also been studied for other viral infections including hepatitis C, dengue, HIV, Japanese encephalitis, norovirus, respiratory syncytial virus, tuberculosis, and West Nile virus. Development has been discontinued for all indications[1][4][6].
