Inaticabtagene autoleucel is an autologous CD19-specific chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (r/r B-ALL). It features a unique single-chain variable fragment (scFv) derived from the HI19α clone, which binds to a distinct high-affinity epitope on CD19 compared to other CAR-T therapies. The construct includes a CD8 hinge and transmembrane domain, 4-1BB costimulatory domain, and CD3 zeta signaling domain. In clinical trials, it demonstrated high overall remission rates and durable responses with manageable safety profiles. It was the first CAR T-cell therapy approved for adult r/r B-ALL in China[1][2][3][4][5][6][9].
