INB-330 is a preclinical, genetically engineered cell therapy developed by IN8bio for the treatment of acute myeloid leukemia (AML). It is part of the DeltEx non-signaling CAR-T (nsCAR-T) platform, which utilizes gamma-delta T cells modified to express a chimeric antigen receptor targeting CD33. Unlike conventional CAR-T therapies that use signaling domains to activate T cells upon antigen binding, the nsCAR construct in INB-330 lacks these domains. This design leverages the innate ability of gamma-delta T cells to distinguish between healthy and malignant tissues based on cellular stress signals, aiming to reduce off-tumor toxicity while maintaining anti-leukemic activity. Preclinical data show that INB-330 can selectively target and kill leukemic cells expressing CD33 while sparing healthy monocytes and hematopoietic stem cells that also express CD33. The approach seeks to address limitations seen with previous CD33-targeted therapies in AML due to toxicity from targeting normal hematopoietic tissue[1][4][6][10].
