Overview

INB-619 is a next-generation gamma-delta (γδ) T cell engager (TCE) developed by IN8bio. It is a bispecific antibody designed to target CD19, a surface antigen broadly expressed on B cells implicated in both hematologic malignancies and autoimmune diseases. Unlike conventional TCEs that activate all T cells via CD3, INB-619 selectively engages γδ T cells—specifically expanding both Vδ1+ and Vδ2+ subsets—to achieve deep, targeted depletion of pathogenic B cells while minimizing inflammatory cytokine release such as IL-6. This approach aims to reduce the risk of severe side effects like cytokine release syndrome (CRS), offering potential for safer immunotherapy in conditions such as systemic lupus erythematosus and B-cell lymphomas. Preclinical data demonstrate potent elimination of disease-causing B cells from lupus patient samples, including those producing harmful IgG1 and IgM antibodies, with significant expansion of γδ T cell populations[1][4][5][7][9].
