Overview

INB-633 is a preclinical, next-generation gamma-delta (γδ) T cell engager (TCE) developed by IN8bio. It is designed to target the myeloid cell surface antigen CD33 and stimulate γδ T cells for the treatment of leukemia, including acute lymphoblastic leukemia (ALL) and acute myeloid leukemia (AML). Unlike conventional CD3-based TCEs that broadly activate all T cells and can cause severe inflammatory side effects such as cytokine release syndrome, INB-633 specifically activates γδ T cells—particularly the Vδ1+ and Vδ2+ subsets—resulting in potent anti-cancer activity with minimal secretion of dangerous cytokines like IL-6. This targeted approach aims to provide deep tumor cell depletion while reducing toxicity risks. The drug has demonstrated strong, dose-dependent killing of leukemia cells in preclinical studies without significant adverse events[1][4][6][8].
