INBRX-109 + pemetrexed is a combination therapy consisting of INBRX-109 (ozekibart), a recombinant humanized tetravalent monoclonal antibody that acts as an agonist of death receptor 5 (DR5), and pemetrexed, an antimetabolite chemotherapy agent. INBRX-109 induces apoptosis in tumor cells through DR5 activation, with favorable safety and anti-tumor activity observed in clinical studies[4][2][5]. Pemetrexed inhibits folate-dependent enzymes essential for DNA synthesis and cell replication. This combination is under investigation primarily for malignant pleural mesothelioma and other metastatic or unresectable solid tumors as part of expansion cohorts in phase 1 studies[1]. The combination aims to enhance anti-tumor efficacy by leveraging synergistic apoptotic and cytotoxic effects.
