INCA-33890 is a bispecific monoclonal antibody under development by Incyte for the treatment of advanced or metastatic solid tumors. It is engineered using Biclonics bispecific antibody technology to simultaneously target and antagonize both programmed cell death protein 1 (PD-1) and transforming growth factor beta receptor 2 (TGFβR2). By binding to PD-1, it blocks the interaction with its ligands PD-L1 and PD-L2, thereby enhancing immune cell-mediated antitumor activity. The drug also selectively inhibits TGFβ signaling in cells co-expressing both targets, aiming to overcome immunosuppression within the tumor microenvironment while minimizing systemic toxicity associated with broad TGFβ pathway inhibition. INCA-33890 is currently in Phase I clinical trials for solid tumors[1][6][8].
