INCA00186 + INCB106385 is an investigational combination therapy consisting of two novel agents developed by Incyte for the treatment of advanced solid tumors, including specific gastrointestinal malignancies (colorectal, gastric/esophageal junction, hepatocellular, pancreatic, and anal cancer) and squamous cell carcinoma of the head and neck. INCA00186 is a humanized, Fc-silenced IgG1 monoclonal antibody that targets and antagonizes CD73, a cell surface enzyme with immunosuppressive properties in the tumor microenvironment, thereby enhancing antitumor immune responses. INCB106385 is a small molecule dual antagonist of the adenosine A2a and A2b receptors, blocking adenosine-mediated immunosuppression. The primary goal of the combination is to overcome tumor-driven immune evasion and improve responses to immunotherapy in cancer patients who are resistant or have progressed after checkpoint inhibitor treatment.
