A combination investigational therapy composed of **INCA00186** (a CD73 inhibitor), **retifanlimab** (a programmed death-1 (PD-1) receptor blocking monoclonal antibody, also known and approved as Zynyz for some indications), and **INCB106385** (a small molecule inhibitor targeting A2A and A2B adenosine receptors); developed by Incyte. This multi-agent regimen is currently under evaluation in phase 1 clinical trials for patients with advanced solid tumors, specifically those with CD8 T-cell-positive cancers such as squamous cell carcinoma of the head and neck and selected gastrointestinal malignancies. The combination is designed to enhance antitumor immunity by simultaneously relieving adenosine-mediated immunosuppression (via CD73 and A2A/B inhibition) and restoring T cell activity through PD-1 blockade[1][3][5].
