Overview

INCA033890 is an investigational, orally bioavailable small molecule inhibitor of cyclin-dependent kinase 2 (CDK2) developed by Incyte Corporation. CDK2 is a serine/threonine kinase that, when complexed with Cyclin E, regulates the G1/S phase transition and S-phase progression of the cell cycle. Dysregulation of the CDK2/Cyclin E pathway, often through CCNE1 amplification, is a known driver of oncogenesis and a mechanism of resistance to CDK4/6 inhibitors in various cancers, including breast, ovarian, and uterine malignancies. INCA033890 is designed to selectively inhibit CDK2, thereby inducing cell cycle arrest and inhibiting the proliferation of tumor cells that are dependent on CDK2 activity. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced or metastatic solid tumors.
