Overview

INCA32459 is a bispecific monoclonal antibody developed by Incyte Corporation in collaboration with Merus. It is designed to simultaneously target two key immune checkpoints: Programmed cell death protein 1 (PD-1) and Lymphocyte activation gene 3 (LAG-3). By blocking these inhibitory pathways, the drug aims to overcome T-cell exhaustion and restore the immune system's ability to identify and eliminate cancer cells. The drug was evaluated in a Phase 1 first-in-human study (INCA32459-101) for patients with advanced solid tumors, including melanoma and PD-L1 positive squamous cell carcinoma of the head and neck. However, in 2024, Incyte announced the discontinuation of the INCA32459 program as part of a strategic pipeline prioritization to focus on other high-impact clinical assets.
