Overview

INCAGN01876 + ipilimumab is a combination investigational immunotherapy pairing INCAGN01876—a glucocorticoid-induced tumor necrosis factor receptor (GITR) agonist monoclonal antibody—with ipilimumab, a cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) blocking monoclonal antibody. INCAGN01876 is designed to activate the immune system by stimulating GITR, leading to the promotion of effector T cell activity and inhibition of regulatory T cells, thereby enhancing anti-tumor responses. Ipilimumab functions as a checkpoint inhibitor by blocking CTLA-4, further promoting T cell activation. This combination is developed for use in patients with advanced or metastatic cancers, and has been evaluated in phase 1/2 clinical trials for safety, tolerability, and preliminary efficacy, including in tumor types such as metastatic solid tumors, endometrial cancer, gastric cancer, and squamous cell carcinoma of the head and neck (SCCHN)[1][2][3].
