Overview

INCB-9471 is an orally available small molecule that acts as a potent, selective, and noncompetitive antagonist of the C-C chemokine receptor type 5 (CCR5). Developed by Incyte Corporation for the treatment of human immunodeficiency virus (HIV) infection, it belongs to a new class of HIV entry inhibitors known as CCR5 antagonists. By binding allosterically to CCR5 at a site distinct from other drugs like maraviroc, INCB-9471 blocks the interaction between HIV-1 gp120 and CCR5 on host cells. This prevents viral entry into CD4+ T cells by inhibiting R5-tropic HIV strains but does not affect CXCR4-tropic or dual-tropic viruses. The drug demonstrated safety and efficacy in reducing viral load during phase I and II clinical trials but did not progress further in development[1][2][3][4][6][7].
