INCB013739 is a small molecule inhibitor of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1), developed by Incyte for the treatment of metabolic diseases such as type 2 diabetes and obesity. By inhibiting 11β-HSD1, the drug reduces the conversion of cortisone to cortisol in tissues, which can improve insulin sensitivity and glycemic control. Clinical studies have shown that INCB013739 improves hepatic and peripheral insulin sensitivity, lowers fasting plasma glucose, total cholesterol, and LDL cholesterol levels in patients with type 2 diabetes. The drug was evaluated as an add-on therapy to metformin in phase 2 clinical trials but has not been approved for any indication[1][2][3][5][6].
