INCB019602 is a small molecule inhibitor of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) that was developed by Incyte for the treatment of type 2 diabetes mellitus. The enzyme 11β-HSD1 is responsible for converting inactive cortisone into active cortisol within specific tissues, notably the liver and adipose tissue. Elevated local cortisol levels contribute to increased hepatic glucose production and insulin resistance. By inhibiting this enzyme, INCB019602 was intended to reduce local cortisol concentrations, thereby improving glycemic control and insulin sensitivity. The compound reached Phase 2 clinical trials, where it was evaluated as an add-on therapy to metformin, but development was ultimately discontinued by Incyte.
