Overview

A combination regimen consisting of the investigational selective Janus kinase 1 (JAK1) inhibitor INCB052793, the selective JAK1 inhibitor itacitinib (also referred to as INCB039110), and the hypomethylating agent azacitidine. INCB052793 and itacitinib inhibit JAK1, reducing signaling in pathways involved in hematologic malignancy pathogenesis. Azacitidine is a nucleoside metabolic inhibitor that induces DNA hypomethylation and cytotoxicity, primarily used in the treatment of myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML)[2][4]. This combination has been studied in clinical trials for relapsed or refractory cases of AML and high-risk MDS, particularly those who have failed prior therapy with hypomethylating agents[2][1][4].
