Overview

INCB098377 is a potent and selective small molecule inhibitor of phosphoinositide 3-kinase gamma (PI3Kγ), currently under development by Incyte Corporation for the treatment of cancer. PI3Kγ is a lipid kinase predominantly expressed in myeloid cells, where it plays a critical role in driving immunosuppressive macrophage programs within the tumor microenvironment. By inhibiting PI3Kγ, INCB098377 facilitates the conversion of M2-polarized macrophages toward a pro-inflammatory phenotype and restores T cell proliferation, thereby enhancing anti-tumor immunity. Preclinical studies have demonstrated that the compound achieves significant tumor growth inhibition in various mouse models at doses of 10 mg/kg without evidence of toxicity, showing efficacy in both inflamed and non-inflamed tumors.
