INCB099280 is a small molecule inhibitor that blocks the interaction of Programmed Death-Ligand 1 (PD-L1) on tumor cells, thereby enhancing immune-mediated tumor cell killing (a PD-L1 inhibitor, a type of immunotherapy)[1][3][7]. Axitinib is a small molecule tyrosine kinase inhibitor that primarily targets the vascular endothelial growth factor receptors (VEGFR-1, VEGFR-2, VEGFR-3), inhibiting angiogenesis (new blood vessel formation required for tumor growth and metastasis). The primary aim of the combination is to synergize immune checkpoint blockade (INCB099280) with inhibition of tumor angiogenesis (axitinib) to enhance anticancer efficacy.
