Overview

INCB106385 is an orally administered small molecule developed by Incyte. It acts as a dual antagonist of the adenosine A2A and A2B receptors, which are involved in immunosuppressive signaling within the tumor microenvironment. By blocking these receptors, INCB106385 aims to enhance anti-tumor immune responses. The drug was investigated as monotherapy and in combination with the PD-1 inhibitor retifanlimab (INCMGA00012) for advanced solid tumors, including non-small cell lung cancer (NSCLC), squamous cell carcinoma of the head and neck (SCCHN), ovarian cancer, castration-resistant prostate cancer (CRPC), triple-negative breast cancer (TNBC), bladder cancer, colorectal cancer (CRC), gastric/gastroesophageal junction cancers, hepatocellular carcinoma (HCC), pancreatic ductal adenocarcinoma (PDAC), and squamous cell anal carcinoma (SCAC). Clinical development included Phase 1 trials focused on safety, tolerability, pharmacokinetics/pharmacodynamics, and preliminary efficacy.
