Overview

INCB123667 + olaparib is an investigational combination regimen consisting of INCB123667, a highly selective small-molecule inhibitor of cyclin-dependent kinase 2 (CDK2), and olaparib, a poly(ADP-ribose) polymerase (PARP) inhibitor. INCB123667 is being studied for its antitumor activity, particularly in cyclin E-overexpressing and platinum-resistant ovarian cancers, where aberrant CDK2 activity drives tumor cell proliferation. Olaparib is approved for various cancers with BRCA1/2 mutations and homologous recombination deficiency, where it disrupts cancer DNA repair. The combination is in clinical development for advanced or metastatic solid tumors, including breast and ovarian cancer, with the aim of targeting tumors through dual cell cycle arrest and DNA damage repair inhibition mechanisms.
