**INCB123667 + ribociclib** is an investigational combination therapy consisting of INCB123667, a potent and selective small-molecule inhibitor of cyclin-dependent kinase 2 (CDK2), and ribociclib, a small-molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). The combination is under clinical evaluation for the treatment of advanced or metastatic solid tumors, particularly HR-positive, HER2-negative breast cancer, with the rationale of dual inhibition of cell-cycle regulatory kinases to enhance anti-tumor efficacy. INCB123667 is being developed by Incyte and ribociclib, approved for certain indications as Kisqali, is developed by Novartis.
