INCB177054

INCB177054 is a novel, potent, orally bioavailable small molecule dual inhibitor of diacylglycerol kinase alpha (DGKα) and zeta (DGKζ), developed by Incyte. It is designed to enhance T-cell activation and lower the threshold for T-cell response to tumor antigens. Preclinical studies show that it synergizes with anti-PD-1 antibodies such as retifanlimab, enhancing anti-tumor immune responses. The drug is currently being evaluated in a phase 1/2 clinical trial as monotherapy or in combination with retifanlimab for patients with select advanced or metastatic solid tumors[3][4][7].
