INCB3344 is a potent, selective, and orally bioavailable small molecule antagonist of the C-C chemokine receptor type 2 (CCR2), developed by Incyte Corporation. It functions by blocking the interaction between CCR2 and its primary ligand, monocyte chemoattractant protein-1 (MCP-1/CCL2), with nanomolar potency. By inhibiting this axis, INCB3344 prevents the migration and infiltration of monocytes and macrophages into tissues, which is a critical process in the pathogenesis of chronic inflammatory diseases. Preclinical research has demonstrated its efficacy in various animal models, including experimental autoimmune encephalomyelitis (a model for multiple sclerosis), inflammatory arthritis, and delayed-type hypersensitivity. It has also been investigated for potential applications in atherosclerosis and choroidal neovascularization.
