Overview

iNCOVACC (BBV154) is a monovalent, replication-deficient chimpanzee adenovirus (ChAd)-vectored intranasal vaccine developed by Bharat Biotech in collaboration with Precision Virologics and Washington University School of Medicine. The vaccine encodes a prefusion-stabilized version of the SARS-CoV-2 spike protein, featuring two proline substitutions in the S2 subunit to enhance immunogenicity. Formulated for intranasal administration via nasal drops, iNCOVACC is designed to elicit both mucosal (IgA) and systemic (IgG and T-cell) immune responses, potentially blocking both infection and transmission. In November 2022, it became the first intranasal COVID-19 vaccine to receive emergency use authorization from India's CDSCO for use as a primary two-dose series and as a heterologous booster dose for adults aged 18 years and above.
