Overview

inCPMV is a UV-inactivated formulation of cowpea mosaic virus (CPMV), a plant virus particle developed as a non-replicating immunostimulatory agent for cancer immunotherapy. CPMV in situ vaccination remodels the tumor microenvironment, activates local and systemic antitumor immune responses, and is already efficacious in several preclinical animal models. inCPMV is rendered non-infectious to plants, addressing agricultural biosafety. Mechanistically, inCPMV signals through TLR2 and TLR4, but not TLR7, due to RNA cross-linking by UV exposure. This mechanism primarily activates the innate immune system. inCPMV is less potent as a monotherapy than wild-type CPMV but demonstrates significantly enhanced and durable antitumor effects when combined with an anti-OX40 agonist antibody in preclinical melanoma models, showing increased tumor infiltration by CD4+ and CD8+ T cells and reduced regulatory T cell presence.
