Overview

Incyclinide is a chemically modified tetracycline, developed to lack antibiotic activity while retaining potent anti-inflammatory and antineoplastic properties. Its primary mechanism of action is the inhibition of matrix metalloproteinases (MMPs), enzymes involved in extracellular matrix degradation, angiogenesis, tumor growth, invasion, and metastasis. By inhibiting MMPs, incyclinide disrupts processes critical for cancer progression and has shown potential to induce apoptosis and tissue necrosis in tumor cells. It has been investigated for various indications including cancers (such as prostate cancer, colon adenocarcinoma, melanoma), HIV-related Kaposi sarcoma, brain tumors, rosacea, acne, inflammatory diseases, cardiovascular disorders, mycoses (fungal infections), periodontal disorders and acute lung injury. Despite promising preclinical results—especially as an inhibitor of tumor invasiveness—clinical trials have not demonstrated efficacy for several cancers or other conditions[1][2][5][6][8].
