Overview

Indatuximab ravtansine is an antibody-drug conjugate (ADC) composed of a chimeric monoclonal antibody targeting CD138 (syndecan-1, SDC1), which is highly expressed on multiple myeloma and some solid tumor cells. The antibody is linked via disulfide bonds to the cytotoxic maytansinoid agent DM4. Upon binding to CD138-positive cells, the ADC is internalized, and DM4 is released inside the cell, leading to cell cycle arrest and apoptotic cell death. Indatuximab ravtansine has demonstrated significant anti-tumor activity in preclinical models of multiple myeloma and triple-negative breast cancer, both as monotherapy and in combination with other agents such as lenalidomide or pomalidomide plus dexamethasone. It remains under clinical investigation for relapsed or refractory multiple myeloma[1][3][5][6].
