Overview

Indibulin is a synthetic small molecule with antimitotic and potential antineoplastic activities. It acts as a tubulin inhibitor, binding to a site on tubulin distinct from those targeted by taxanes or Vinca alkaloids, leading to destabilization of microtubule polymerization. This results in tumor cell cycle arrest at the G2-M phase and induction of apoptosis. Notably, indibulin has demonstrated activity against multidrug-resistant and taxane-resistant tumor cell lines and lacks the neurotoxicity typically associated with other tubulin-binding agents. It is orally bioavailable and has been investigated primarily for advanced solid tumors, including metastatic breast cancer[1][3][5].
