Overview

Individual Patient TCR-Transduced PBL is an investigational autologous T-cell therapy developed by the National Cancer Institute (NCI) Surgery Branch. The therapy consists of a patient's own peripheral blood lymphocytes (PBL) that have been genetically modified using gamma-retroviral vectors to express T-cell receptors (TCRs) specific to neoepitopes derived from mutations in the TP53 or RAS (KRAS and NRAS) genes. These mutations are prevalent in various hematologic malignancies, including acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), and multiple myeloma. The therapeutic process involves the isolation of patient T cells via leukapheresis, ex vivo stimulation and retroviral transduction, and subsequent re-infusion into the patient following lymphodepleting chemotherapy. The engineered T cells are designed to recognize and eliminate cancer cells presenting these specific mutant neoantigens in the context of the patient's HLA molecules.
