IPX (Individualized Peptide Vaccine) is a personalized cancer vaccine developed by the University Hospital Tuebingen for the treatment of pediatric and adolescent/young adult (AYA) patients with metastasized fusion-driven sarcomas, including Ewing sarcoma, rhabdomyosarcoma, and synovial sarcoma. The vaccine is tailored to each patient by identifying sarcoma-specific peptides through next-generation whole exome sequencing (WES) and RNA sequencing of both tumor and healthy tissue. These patient-specific peptides are formulated with the adjuvant XS15 (a TLR1/2 ligand) to stimulate a robust cytotoxic T-cell response against the tumor. This approach is designed to be applicable regardless of the patient's HLA-typing or the expression of common tumor-associated antigens. It is currently being evaluated in the Phase I/II PerVision trial (NCT04416568).
