Overview

indocyanine green + p28 (ICG-p28) is an intraoperative near-infrared (NIR) fluorescence imaging probe designed for the real-time identification of tumor margins during surgery. It consists of the fluorescent dye indocyanine green (ICG) conjugated to p28, a 28-amino acid cell-penetrating peptide derived from the bacterial protein azurin. The p28 peptide serves as a tumor-targeting moiety that preferentially enters cancer cells and binds to the DNA-binding domain of the p53 tumor suppressor protein, though its primary role in this conjugate is to facilitate selective accumulation of ICG within malignant tissues. This targeted approach is intended to improve the accuracy of breast-conserving surgery for ductal carcinoma in situ (DCIS) and invasive breast cancer by clearly delineating tumor boundaries, thereby reducing positive margin rates and the necessity for follow-up re-excision procedures.
