Indole-3-carbinol (I3C) is a naturally occurring compound derived from the breakdown of glucobrassicin, a glucosinolate found in cruciferous vegetables such as broccoli, Brussels sprouts, cabbage, cauliflower, kale, and turnips. It is also available as a dietary supplement. In the acidic environment of the stomach, I3C undergoes condensation to form several biologically active products including diindolylmethane (DIM). Preclinical studies suggest that I3C and its metabolites modulate biotransformation enzymes involved in hormone metabolism and detoxification pathways. Mechanistically, it exhibits anti-estrogenic effects by altering estrogen metabolism—shifting it toward less potent forms—and may inhibit cell proliferation and induce apoptosis in cancer cells. It has been studied for potential roles in cancer prevention (especially hormone-dependent cancers), cervical dysplasia related to HPV infection, recurrent respiratory papillomatosis, liver protection, and immune modulation. However, clinical evidence for efficacy remains limited; some studies suggest possible chemopreventive effects while others indicate potential risks or tumor-promoting activity under certain conditions[1][2][4][5].
