Overview

Indoprofen is a nonsteroidal anti-inflammatory drug (NSAID) that was originally developed and marketed for its analgesic and anti-inflammatory properties. It acts primarily as an inhibitor of prostaglandin G/H synthase 1 and 2 (COX-1 and COX-2), thereby reducing the synthesis of prostaglandins involved in inflammation and pain. Indoprofen also inhibits C-X-C chemokine receptor type 1. The drug was withdrawn worldwide in the 1980s due to reports of severe gastrointestinal bleeding and concerns about carcinogenicity observed in animal studies. Beyond its original indications, research has shown that indoprofen can increase production of survival motor neuron protein via a cyclooxygenase-independent mechanism, suggesting potential utility for spinal muscular atrophy; it has also been shown to activate the PDK1/AKT/S6K pathway in muscle cells, indicating possible benefit against muscle wasting conditions[3][5][6][7].
