**Indoximod + nivolumab** is an investigational combination therapy for unresectable or metastatic melanoma and other advanced cancers. Indoximod is a small molecule inhibitor targeting the indoleamine 2,3-dioxygenase (IDO) pathway, designed to block the ability of cancer cells to suppress immune response via IDO, thereby enhancing antitumor immunity. Nivolumab is a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) immune checkpoint, reactivating T-cell-mediated immune response against tumor cells. The combination was evaluated in a Phase II/III randomized trial, where indoximod is given concomitantly with nivolumab to potentially improve checkpoint inhibitor efficacy, especially in malignant melanoma.
