Overview

**Indoximod + nivolumab** is an investigational combination therapy for unresectable or metastatic melanoma and other advanced cancers. Indoximod is a small molecule inhibitor targeting the indoleamine 2,3-dioxygenase (IDO) pathway, designed to block the ability of cancer cells to suppress immune response via IDO, thereby enhancing antitumor immunity. Nivolumab is a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) immune checkpoint, reactivating T-cell-mediated immune response against tumor cells. The combination was evaluated in a Phase II/III randomized trial, where indoximod is given concomitantly with nivolumab to potentially improve checkpoint inhibitor efficacy, especially in malignant melanoma. Indoximod remains experimental in this combination, while nivolumab is approved as monotherapy and with other agents for various cancers[6][7][9].
