**Indoximod + paclitaxel** is a combination regimen under investigation for metastatic breast cancer and other solid tumors. Indoximod is a small molecule inhibitor of the indoleamine 2,3-dioxygenase (IDO) pathway, designed to reverse tumor-induced immunosuppression by blocking IDO-mediated tryptophan metabolism, restoring T and NK cell function and enhancing anti-tumor immunity[6]. Paclitaxel is a microtubule-stabilizing chemotherapeutic agent that induces apoptosis in dividing cancer cells. Preclinical studies suggested synergy between indoximod and taxanes such as paclitaxel due to increased effector T cells at the tumor site[6]. Clinical trials have evaluated the efficacy and safety of this combination, particularly in ERBB2-negative metastatic breast cancer[1][2]. The combination did not show statistically significant improvement in progression-free survival in phase II trials but remains of interest for investigative immunotherapy approaches[1][2].
